Product DescriptionThis product is designed to detect the activity for variety of kinases (see Table 1) and screening the inhibitors of the corresponding kinase. The assay kit is non-radioactive but colorimetric.
The principle of the assay is that:
The kinase substrates is immobilized onto the 96-well plate. The kinases use ATP to phosphorylate the substrate on the plate. The phosphorylated product is detected by the phosphor-substrate specific antibodies. See figure1 for the principle illustration.

測試盒原理:

 基于ELISA的基本原理。PKC底物被包被在酶標板上,在ATP存在的前提下,PKC激酶將底物磷酸化,洗掉游離的PKC激酶和其他成分。再加入特異性磷酸化抗體(只識別磷酸化底物,而與非磷酸化底物無交叉反應。)檢測殘留在酶標板上的磷酸化底物。通過酶標二抗放大信號,讀出OD值(蛋白激酶活性與OD值的對數成正比)。與對照組相比,得出處理組的蛋白激酶活性。
